Brief Summary in Scientific Language

Temporal lobe epilepsy is characterized by epileptic seizures of mostly hippocampal origin and structural changes in the hippocampus. These include the loss of principal neurons and interneurons in parts of the Ammon's horn (CA3, CA1) and in the hilus, reactive gliosis and granule cell dispersion. In contrast, neurons of the CA2 region are mostly resistant against pathological changes. Still, the function of this region is only rarely investigated. During the last years new antibodies and cDNA probes have been developed to better investigate the CA2 region and its connectivity. We want to use these methods to better characterize the survival of CA2 neurons and their synaptic integration in the hippocampal neuronal network in human specimens from patients with intractable temporal lobe epilepsy. Furthermore we want to do electrophysiological recordings in slices to investigate epileptogenic changes on the single cell level. The understanding of the function of the CA2 region for the onset and propagation of epileptic seizures in temporal lobe epilepsy might besides the novel information for basic science further lead to new therapeutic options, e.g. through deep brain stimulation. The latter are interesting since temporal lobe epilepsy is mostly pharmacoresistant and since the surgical therapy cannot be applied if there is a bilateral focus.
